Near-infrared Raman Spectroscopic Measurements of Salmon Meat Composition
Biological tissues are a complex, heterogeneous material. Proteins and fats represent the major constituents in tissue, with minor components such as cells, polysaccharides, small molecules, and pigments. Understanding the chemical properties of biological tissues has important implications in biomedical and food meat quality applications. In biomedical applications, Raman has been used extensively to improve our understanding of chemical changes in tissue associated with disease or aging status. In food meat quality applications, Raman has been used in the laboratory as a quality assurance tool, to characterize hydrolyzed products, and to identify major contaminants. In this study, we describe how the use of 785 nm or 1000 nm excitation can provide rapid Raman spectroscopy measurements in salmon meat. Store-bought specimens of wild-caught, farmed, and smoked salmon were examined by Raman spectroscopy. Spectral bands were compared against literature references for collagen, carotenoids, and salmon tissue. Raman spectra of salmon tissue reflected the spatial heterogeneity of the salmon tissue. Spectral contributions from collagen protein, fats, and carotenoid-based pigments were observed. Finally, we describe fiber optic sampling strategies for direct tissue and transcutaneous measurements.
